生化机理 | Fibronectins bind cell surfaces and various compounds including collagen, fibrin, heparin, DNA, and actin. Fibronectins are involved in cell adhesion, cell motility, opsonization, wound healing, and maintenance of cell shape. Involved in osteoblast compaction through the fibronectin fibrillogenesis cell-mediated matrix assembly process, essential for osteoblast mineralization. Participates in the regulation of type I collagen deposition by osteoblasts. Anastellin binds fibronectin and induces fibril formation. This fibronectin polymer, named superfibronectin, exhibits enhanced adhesive properties. Both anastellin and superfibronectin inhibit tumor growth, angiogenesis and metastasis. Anastellin activates p38 MAPK and inhibits lysophospholipid signaling. Post-translational: Sulfated. It is not known whether both or only one of Thr-2155 and Thr-2156 are/is glycosylated. Forms covalent cross-links mediated by a transglutaminase, such as F13A or TGM2, between a glutamine and the epsilon-amino group of a lysine residue, forming homopolymers and heteropolymers (e.g. fibrinogen-fibronectin, collagen-fibronectin heteropolymers). Phosphorylated by FAM20C in the extracellular medium. Proteolytic processing produces the C-terminal NC1 peptide, anastellin. Some lysine residues are oxidized to allysine by LOXL3, promoting fibronectin activation and matrix formation. Serotonylated on Gln residues by TGM2 in response to hypoxia. |
产品介绍 | 纤连蛋白(Fibronectin),是细胞外基质中的一种高分子量(~450kDa)糖蛋白,由2个分子质量约220kDa的亚基通过链间二硫键连接而成,广泛分布于机体组织中。在生命科学研究中,具有广泛的应用,比如作为细胞培养基质,提高细胞贴壁率,增强细胞代谢水平,缩短细胞生长时间,另外也可以直接添加至培养基中,促进细胞生长,增殖、维持良好细胞状态。 OsrhFN是一种来源于水稻胚乳细胞的重组人纤连蛋白,相比动物提取来源的纤连蛋白而言,无动物源性,无潜在病毒感染风险,更好的批次稳定性。 pH值: 6.0-8.0 产品应用: 细胞培养,体外诊断,实验检测,美妆等 使用方法 1、将OsrhFN用PBS 稀释至终参考浓度范围50-100μg/ml,混匀后,加入到孔板或培养瓶中,使OsrhFN液铺满培养瓶/板底。具体OsrhFN添加量参考表1。 2、将加有OsrhFN的培养瓶/板,4℃过夜或者37℃包被1小时; 3、吸净培养瓶/板中的OsrhFN液,收集后请置于4℃保存,1周内可用于其他培养瓶/板包被; 4、取出OsrhFN包被后的培养瓶/板,直接接种细胞即可; 5、如包被后的培养瓶/板暂不使用,请置于4℃密封保存,建议一周内用完。 B、细胞培养: 直接加入细胞培养基中,使用终浓度参考范围5-50ug/ml。 Fibronectin (FN) is a particularly important and well-studied component of the extracellular matrix, and is known to play a key role in cell adhesion, growth, spreading, migration, differentiation and proliferation. Fn is a 200-250 kDa glycoprotein composed of 2 subunits bound via a disulfide bond. Currently, the Fn is purified from the plasma, which however is limited by the availability of supply. The the recombinant human fibronectin (OsrhFN) was expressed in the rice endosperm platform, which is animal component free and has high purity, and has been demonstrated has the same physical and chemical with the plasma derived Fn. OsrhFN provides a safety solution to replace the plasma derived FN. |
